Scutellarein ameliorates inflammation and oxidative stress in L-arginine induced acute pancreatitis in mice through regulating the expression of VEGF

Abstract

Background

Acute pancreatitis (AP) is an abdominal disease mainly characterized by oxidative stress and inflammation of the pancreas. Studies have reported that scutellarein has antioxidant and anti-inflammatory properties.

Objectives

To explore whether scutellarein is protective against AP induced by L-arginine.

Results

Scutellarein administration mitigated inflammatory infiltration, edema and acinar cells necrosis of pancreatic tissue, and dramatically reduced the elevated serum levels of amylase in AP mice. Scutellarein inhibited the activity of MPO, MDA and ROS, and inhibited VEGF expression and the phosphorylation of NF-kBp65 in pancreatic tissues of AP mice, and reduced the production of pro-inflammatory cytokines Interleukin 6 (IL-6), Tumor necrosis factor α (TNF-α) and IL-1β. In addition, through AAV-VEGF upregulated the expression of VEGF in AP mice, the protective effect of scutellarein on AP mice were reversed.

Conclusion

Scutellarein treatment alleviated pancreas injury of AP mice by inhibiting oxidative stress and inflammatory response via downregulating VEGF expression.
