1. 利用小鼠多能干细胞神经分化并结合鸡胚神经发育模型,阐明不同信号通路在神经诱导和分化中的功能及其调控机制;揭示表观遗传调控网络与细胞外信号在多能干细胞神经命运决定中的功能及其相互调控关系。 2. 利用单细胞测序技术体系,建立小鼠早期胚胎的分子表达图谱,筛选并鉴定外胚层干细胞的标记基因,建立小鼠胚胎外胚层发育分化的细胞谱系。 3. 建立人多能干细胞神经定向分化模型,筛选并鉴定人多能干细胞神经分化不同阶段的标记基因,阐明人多能干细胞神经分化的分子机制;诱导人多能干细胞分化为具有特定功能的神经元,探索神经退行性疾病干细胞治疗的可能性。
